Effects Of Phthalates On Motility Behavior Of Bull Sperm Cells

...behaviour of reactivated deep frozen bull sperm cells were investigated. A total of twenty experiments were carried out for this work to be successful. Four of them were control experiments and sixteen were the test or impact experiments. In the control experiments, the sperm cell suspension was a 1:2 dilution of reactivated bull semen and 300 mosmol HEPES basic medium. While in the impact experiments, HEPES basic medium was changed to HEPES basic plus stress compound. Four different concentrations of Dimethyl Phthalates and Dibutly Phthalates were used as stress compounds. Motility parameters were measured under exposure of bull sperm cells to 10-3, 10-4, 10-5and 10-6 molar solutions of DMP and 10-5, 10-6, 10-7 and 10-8 molar solutions of DBP. It was also possible to compare the effects of both DMP and DBP at the same concentrations of 10-5 and 10-6 mol/l DMP and DBP.
A Computer Assisted Sperm Analysis (CASA) system was used for measurement of motility parameters relevant to the study such as; Average Path Velocity (VAP), Straight Line Velocity (VSL), Curvilinear Velocity (VCL), Percentage Motility, Amplitude of Lateral Head Displacement (ALH), Head Cross Beat Frequency (BCF) and derived parameters of motility such as Linearity, Straightness and Wobble. The data derived from the CASA measurement were exported directly to a Microsoft Excel file for data analysis and the results from the impact experiments were compared with that from the control experiments.
DMP was found to be negatively associated with the percentage of motile sperm cells, while there was no consistent relationship between DBP and the percentage of motile sperms after exposure for 120 minutes, and no dose response relationship was found for any of the compounds.
